From Austin:
Take highway 290 West from Austin, through Johnson City and toward Fredericksburg. Grape Creek Vineyards is on your left approximately 20 miles past Johnson City and 10 miles before you reach Fredericksburg.
From San Antonio:
Take I-10 West from San Antonio. Take Highway 87 North at the Comfort Exit and head North to Fredericksburg (20 miles from exit) then take main street (turns into Highway 290) East for 10 miles to Grape Creek Vineyards.
Another option from San Antonio is to take 1376 North after exiting from I-10. It is a more scenic country road and a little shorter but requires a slower pace.
